木榄是一种具有较高经济价值的药用红树植物,对沿海地区的生态也有良好的保护作用。本研究通过提取海南木榄叶的DNA,再利用紫外光谱和荧光光谱法对两种重金属离子Pb2+和Cd2+与海南木榄叶DNA的相互作用进行了研究。结果表明,Pb2+和Cd2+的存在对木榄叶DNA的结构有一定影响,且均与以沟槽结合的模式与木榄叶DNA相互作用;热力学参数表明Pb2+与木榄叶DNA可自发反应,氢键和范德华力为主要的相互作用力,而Cd2+与DNA为非自发反应,其相互作用力以疏水作用力为主。该研究可为深入了解红树植物从耐重金属离子的角度能保护生态的作用机制提供理论支持。 |

Bruguiear gymnorrhiza is one kind of medicinal mangrove plants with higher commercial value, which also has well protective effect to the ecology of the coastal area. In the present study, DNA is extracted from the leafs harvested from Hainan Bruguiear gymnorrhiza. Then the interaction of two kinds of heavy metals including lead ion,cadmium ion to the DNA were been investigated by using UV-vis and fluorescence spectroscopes. The results indicated that the presence of lead ion and cadmium ion had a certain influence on the structure of DNA and the interaction could occur in a groove-binding mode. The thermodynamic parameters showed that it is a spontaneous reaction between lead ion and DNA with main interaction forces as hydrogen bond and Vander Waals force. However, cadmium ion unspontaneously interacted with DNA and hydrophobic force played a main role in the binding process. This research can provide some theoretical support for further understand on the mechanism of action of mangrove plants protecting ecology from the point of view its tolerance of heavy metal ions. |
