      基于网络药理学探讨醒神解郁汤治疗卒中后抑郁的分子作用机制,并通过细胞实验进 行相关生物学事件验证。通过TCMSP、化源网、Chemistry Database等数据库及相关文献收集醒神 解郁汤中单味药的化学成分,结合口服利用度(OB)≥30%、类药性(DL)≥0.18及Swiss数据库筛选出 活性成分并预测其靶点,构建中药-成分-靶点网络图。筛选活性成分与疾病交集核心靶点,构建 蛋白互作网络图并进行拓扑分析。通过Metascape数据库进行KEGG通路和GO功能富集分析。基 于网络药理学分析获得通路及核心靶点,建立神经元细胞、星形胶质细胞糖氧剥夺模型,并检测细 胞存活率。筛选出醒神解郁汤共有成分有 128 个,共有靶点 460 个,交集核心靶点 57 个。其中 AKT1与32个靶点发生相互作用,PI3K-Akt信号通路可能是醒神解郁汤治疗卒中后抑郁的关键作 用信号通路。MTT实验结果显示醒神解郁汤能够有效提高神经元细胞、星形胶质细胞在缺血缺氧 条件下的存活率。醒神解郁汤能通过核心靶点治疗卒中后抑郁。

      This study aimed to explore the molecular mechanism of Xingshen Jieyu Decoction in the treatment of poststroke depression based on network pharmacology, and to verify the relevant biological events through cell experiment. The chemical components of single drug in Xingshen Jieyu Decoction were collected through tcmsp, huayuan.com, chemistry da‑ tabase and other databases and relevant literature. Combined with oral availability (OB)≥30%, drug class (DL)≥ 0.18 and Swiss database, the active components were screened and their targets were predicted, and the traditional Chinese medicine component target network diagram was constructed. The core targets of the intersection of active components and diseases were screened, the protein interaction network was constructed and the topological analysis was carried out. The KEGG pathway and go function enrichment analysis were carried out through metascape database. Based on the pathways and core targets obtained by network pharmacological analysis, the oxygen glucose deprivation models of neurons and astrocytes were established, and the cell survival rate was detected. Results showed that there were 128 components, 460 targets and 57 intersection core targets in Xingshen Jieyu Decoction. AKT1 interacted with 32 targets, and PI3K Akt signal pathway might be the key signal pathway of Xingshen Jieyu Decoction in the treatment of post-stroke depression. MTT test results showed that Xingshen Jieyu Decoction could effectively improve the survival rate of neurons and astrocytes under ischemia and hypoxia. Xingshen Jieyu Decoction could treat post-stroke depression through its core target.
