      为了观察黄芩和苍术对湿阻中焦证证候模型燥湿功效的差异,阐明黄芩和苍术燥湿作 用与药性组合的关联性,将64只雄性SD大鼠随机分为正常组、模型组、黄芩高剂量组、黄芩中剂量 组、黄芩低剂量组、苍术高剂量组、苍术中剂量组和苍术低剂量组。正常组常规喂养,其余各组通 过饱时内服油脂、饥时灌胃冰水和外部寒湿环境作用20 d复制湿阻中焦证模型。采用苏木素-伊 红(HE)染色观察大鼠胃和结肠组织的病理学变化,采用胃液游离酸度测定方法检测胃液酸度,采 用胃蛋白酶活性测定方法测定胃蛋白酶活力,使用蛋白免疫印迹法(Western blot)检测大鼠结肠组 织水通道蛋白3(AQP3)表达情况。结果表明,与正常组比较,模型组大鼠宏观表征显示具有湿阻 中焦证的表现,胃、结肠组织有炎症反应,胃液游离酸度降低(P<0.05),胃蛋白酶活力减弱(P< 0.05);结肠组织中AQP3蛋白表达量有所下降。与模型组比较,给药组大鼠宏观表征得到改善,黄 芩和苍术对模型大鼠胃和结肠组织炎症都有一定的改善作用,苍术各剂量组均可升高模型大鼠胃 液游离酸度(P<0.05)和增强模型大鼠胃蛋白酶活力(P<0.05),黄芩各剂量组也可升高模型大鼠胃 液游离酸度和增强模型大鼠胃蛋白酶活力,但差异无统计学意义,黄芩中、低剂量组和苍术高、中、 低剂量组均能显著降低结肠组织AQP3蛋白表达,研究证明黄芩和苍术对湿阻中焦证大鼠的燥湿 作用与其药性组合具有一定的关联性,即其燥湿功效的强弱与其寒热药性有关,苦温的苍术比苦 寒的黄芩作用效果更好。

      To observe the difference of dryness-dampness effect of Scutellaria baicalensis and Rhiizoma Atractylodis on the model of syndrome of dampness hampering middle jiao, and to clarify the relationship between the dryness-dampness effect of them and the combination of medicinal properties. Sixty-four male SD rats were randomly classified into the nor‑ mal group, model group, Scutellaria baicalensis high-, medium-, and low-dose groups, as well as Rhizoma Atractylodis high-, medium-, and low-dose groups. The normal group was fed routinely, and the other groups were fed with fat during satiety, ice water during hunger and external cold and humid environment for 20 days to replicate the model of syndrome of dampness hampering middle jiao. The macroscopic characterization of rats was observed, the pathological changes of stom‑ ach and colon were observed by hematoxylin-eosin (HE) staining, the acidity of gastric juice was measured by free acidity of gastric juice, the activity of pepsin was determined by pepsin activity assay, and the expression of aquaporin 3 (AQP3) in rat colon was detected by Western blotting (Western blot). The results showed that compared with the normal group, the macroscopic characterization of the model group showed that the rats in the model group had the syndrome of dampness hampering middle jiao. There were inflammatory reactions in stomach and colon, the concentration of free acidity in gastric juice decreased, the activity of pepsin decreased, and the expression of AQP3 protein in colon decreased. Compared with the model group, the macroscopic characterization of rats in the treated group was improved, and Scutellaria baicalensis and Rhizoma Atractylodis could improve the inflammation of stomach and colon tissue of model rats to a certain extent. Atracty⁃ lodes macrocephala could increase the concentration of free acidity of gastric juice (P<0.05) and enhance the activity of pepsin in model rats (P<0.05). All dose groups of Scutellaria baicalensis could also increase the concentration of free acidity of gastric juice and enhance the activity of pepsin in model rats, but the difference was not statistically significant. Scutellaria baicalen⁃ sis medium and low dose groups and all dose groups of Rhizoma Atractylodis could significantly reduce the expression of AQP3 protein in colon tissue of model group. Research showed that the dryness-dampness effect of Scutellaria baicalensis and Rhi⁃ zoma Atractylodis macrocephala on rats with syndrome of dampness hampering middle jiao was related to their combination of medicinal properties. Specifically, the efficacy of dryness-dampness was related to their cold/heat property, and the resulting outcome of bitter-warm Rhizoma Atractylodis was better than that of bitter-cold Scutellariae baicalensis.
