      以一株浙江近无柄雅榕古树为研究对象,通过Illumina HiSeq测序平台对其叶绿体基 因组进行了测序,分析了其叶绿体基因组结构特征及其近缘种间关系。结果表明:近无柄雅榕叶 绿体 DNA 全长 160 292 bp,具有典型的环状结构,由 4 个区域组成,包括 1 个大单拷贝区(large single copy,LSC)、1 个小单拷贝区(small single copy,SSC)和一对分开的反向重复区域(inverted repeat,IR),LSC、SSC、IR的长度分别为88 565、20 145和25 791 bp;共注释出118个基因,其中包括 4个rRNA基因、31个tRNA基因和83个蛋白质编码基因;共检索到68个SSR,其中包括11个复合型 SSR、52个单碱基重复、4个二碱基重复和1个三碱基重复;与蛋白质编码基因对应的密码子偏好使 用A/T碱基,在所有密码子中编码亮氨酸(Leu)的密码子最多;在系统发育树上,近无柄雅榕与已报 道的菩提树亲缘关系最近,同源性达99%以上。

      The complete chloroplast genome of Ficus concinna Miq. var. subsessilis Corner sequenced by Illumina HiSeq sequencing platforms was analyzed and compared with the reported chloroplast genomes of genetically close species. The results showed that it was a circular molecule of 160 292 bp in length with a typical quadripartite structure, including a pair of inverted repeats (IR) of 25 791 bp that were separated by large and small single copy regions (LSC and SSC) of 88 565 bp and 20 145 bp, respectively. 118 chloroplast genes were successfully annotated, including 4 rRNA genes, 31 tRNA genes and 83 protein-coding genes. A total of 68 SSR were identified, including 11 compound SSR, 52 mononucleotides, 4 dinu⁃ cleotides and 1 trinucleotides. The codon usage of protein-coding genes was biased touse A/T bases. Moreover, among these codons, leucine were the most used amino acids. The phylogenetic analysis showed that it had a closer genetic rela⁃ tionship with reported Ficus religiosa (homology of 99%).
