      通过2,3,5-氯化三苯基四氮唑(TTC)染色法,探究5 种热带林木种子生活力最佳染色条 件。结果表明,影响5 种热带林木种子染色的主要因素为预处理时间、染色温度与TTC 浓度的互作 效应、染色温度等。5 种林木种子的最佳染色条件不同。榄仁树种子生活力指数较高,受试验条件 的影响不大,可选取任意一组试验条件作为最佳染色条件。水黄皮种子的最佳染色条件为:染色 温度35 ℃,TTC 浓度1.0%,温水浸泡时间12 h。银合欢种子的最佳染色条件为:染色温度35 ℃, TTC 浓度0.5%,热水浸泡时间96 h。见血封喉种子的最佳染色条件为:染色温度40 ℃,TTC 浓度 1.0%,温水浸泡时间36 h。黄槿种子的最佳染色条件为:染色温度30 ℃,TTC 浓度0.5 %,硫酸浸泡 时间1 h。研究结果可为基于种子生活力筛选天然更新能力较强的海防林树种提供依据。

      Based on 2, 3, 5-Triphenyl Tetrazolium Chloride(TTC)method, the optimum staining conditions of seed via⁃ bility for five species of tropical trees were studied. The results showed that the main factors affecting the staining of seeds for different species of trees were different. The pretreatment time was the most important factor affecting the staining re⁃ sult of various seeds, followed by the interaction effects between the staining temperature and the TTC concentration, and staining temperature. The optimum staining conditions of different tree species were different. The viability of Terminalia catappa was higher, which was not affected by the experimental conditions, therefore, any group of experimental conditions can be seleted as the test staining conditions. The optimum staining conditions of Pongamia pinnata were: the staining tem⁃ perature 35℃, TTC concentration 1.0% and immersion times in warm water 12 h. The optimum staining conditions of Leu⁃ caena leucocephala were: the staining temperature 35℃, TTC concentration 0.5% and immersion times in hot water 96 h. The optimum staining conditions of Antiaris toxicaria were: the staining temperature 40℃, TTC concentration 1.0% and im⁃ mersion times in warm water 36 h. The optimum staining conditions of Hibiscus tiliaceus were: the staining temperature 30℃, TTC concentration 0.5% and immersion time in sulfuric acid for 1 h. It could be used to select tree species of coastal windbreaks with strong regeneration ability, based on seed viability.
