对栽培胆木药材的性状进行鉴定,确立胆木最适种植环境和采收年限. 采集海南省10个乡镇种植的胆木、五指山市水满乡不同株龄的胆木和野生胆木,利用显微鉴别、显色反应和薄层鉴别鉴定药材性状,利用HPLC 法测定异常春花苷内酰胺含量,分析不同产地、不同株龄的胆木中异常春花苷内酰胺的含量差异. 结果显示:栽培胆木的药材性状、显微性状、药材粉末的显色反应、薄层显色反应等均与野生药材相符;峰面积与异常春花苷内酰胺浓度的回归方程为Y = 73843X+76394(R2 = 0.9999),来自10 个乡镇的胆木中异常春花苷内酰胺的质量百分比含量(%)为:南圣镇1.355,水满乡1.97,番阳镇1.03,毛感镇2.052,什玲镇2.097,响水镇1.96,和平镇2.453,乌什镇1.86,长征镇1.26,红毛镇2.015. 种植于水满乡株龄2~6 年的胆木中异常春花苷内酰胺的含量分别为(%):0.472、0.876、1.690、1.936、1.783,而野生胆木异常春花苷内酰胺含量为1.679%. 因此,环境对胆木中异长春花苷内酰胺的积累有显著影响,种植时应选择适宜环境;株龄4 年及其以的胆木质量达到海南省地方标准,方可入药. |

This research is to identify the Nauclea officinalis Pierre ex Pitard cultivated in plantion and make clear the optimum environment for planting and the suitable harvest period. The material of Nauclea officinalis Pierre ex Pitard is collected in plantation in ten towns of four counties of Hainan province, and the materials with different ages are from Wuzhishan county of Hainan province. The properties of medicine material are identified by the technologies of microholography, color reaction and thin-layer chromatography. The contents of isovincoside lactone in medicinal materials are detected by HPLC to differentiate between superior and inferior materials. The results of microholography, color reaction and thin-layer chromatography of materials from plantation are the same to those of the wild; the equation is figured out according to the results of peak area and the contents of isovincoside lactone. The isovincoside lactone mass concentrations in different materials from ten townships are as follows: Nan Sheng 1.355%, Shui Man 1.97%, Fan Yang 1.030%, Mao Gan 2.052%, Shen Ling 2.097%, Xiang Shui 1.96%, He Ping 2.453, Wu Shen 1.86%, Chang Zheng 1.26%, Hong Mao 2.015. The mass concentrations of isovincoside lactone in materials with 2~6 ages are respectively 0.472%, 0.876%, 1.690%, 1.936% and 1.783%, and that in wild material is 1.679%. The content of isovincoside lactone is affected remarkably by the environment, so this species is cultivated in suitable plantation. The materials from the trees with 4 ages are up to the prescribed standard of Hai nan province. |
