| 线粒体作为细胞内重要的细胞器之一,其粘度的异常变化与多种疾病的发展密切相关,为此本研究设计并合成了两种基于吩噻嗪-喹啉盐的D-π-A型荧光探针PTO1和PTO2,用于监测线粒体粘度变化。该探针以吩噻嗪为电子供体(D),喹啉盐作为电子受体(A),通过π桥进行连接,其均展现出了优异的光物理性能。在一定的粘度范围内,二者荧光强度与粘度呈现出良好的线性关系,并表现出高抗干扰性、较强的光稳定性以及低细胞毒性。其中,PTO1能够不依赖线粒体膜电位靶向在线粒体上。进一步细胞实验表明,PTO1可有效响应制霉菌素(Nys)、过氧化氢(H2O2)及脂多糖(LPS)刺激所引起的线粒体粘度的变化。因此,探针PTO1有望作为一种检测细胞粘度变化的优良工具。 |

| As one of the important organelles within cells, mitochondria are closely related to the development of various diseases when their viscosity changes abnormally. In this study, two D-π-A type fluorescent probes PTO1 and PTO2 based on phenothiazine-quinoline salts were designed and synthesized to monitor changes in mitochondrial viscosity. These probes use phenothiazine as the electron donor (D) and quinolinium as the electron acceptor (A), connected through a π-bridge, and both exhibit excellent photophysical properties. Within a certain viscosity rang, the fluorescence intensity of both probes showed a good linear relationship with viscosity, along with high anti-interference capability, strong photostability, and low cytotoxicity. Among them, PTO1 could target mitochondria independently of membrane potential. Further cellular experiments indicated that PTO1 could effectively respond to changes in mitochondrial viscosity induced by nystatin (Nys), hydrogen peroxide (H2O2), and lipopolysaccharide (LPS) stimulation. Therefore, probe PTO1 is expected to serve as an excellent tool for detecting changes in cellular viscosity. |
