线粒体是细胞内重要的能量提供者,参与细胞代谢和信号传递等关键生物过程。当线粒体功能异常时,可能导致细胞功能紊乱,严重时会引发多种疾病。线粒体粘度的变化是评估线粒体功能的重要指标,具有重要的研究意义。基于这一需求,以苯并噻唑衍生物为核心结构,引入吡啶盐和喹啉盐,设计合成了三种具有D-π-A构型的荧光探针BTZ1-BTZ3。并进一步研究了探针的光物理性质及生物学性质。实验结果表明,探针BTZ1和BTZ3对溶液粘度变化具有较好的响应,所有探针均表现出良好的光稳定性和较低的细胞毒性,而且探针BTZ3能够有效监测线粒体内的粘度变化。 |

Mitochondria serve as critical energy producers within cells and play a pivotal role in essential biological processes, including cellular metabolism and signaling pathways. Dysfunction of mitochondria can result in cellular impairment, potentially leading to a wide range of diseases. Mitochondrial viscosity is a key indicator for assessing mitochondrial function and holds significant research value. To address this need, three D-π-A type fluorescence probes (BTZ1-BTZ3) were designed and synthesized, utilizing benzothiazole derivatives as the core structure and incorporating pyridinium and quinolinium salts. The photophysical and biological properties of these probes were thoroughly investigated. The findings revealed that BTZ1 and BTZ3 exhibited excellent responsiveness to viscosity changes, while all probes demonstrated superior photostability and minimal cytotoxicity. Notably, probe BTZ3 effectively monitored viscosity alterations within mitochondria. |
